Il Corso è di 8 crediti (64 ore). 7 crediti sono dedicati alla didattica
frontale, attraverso diapositive power point che saranno rese disponibili
agli studenti, e un credito è dedicato a seminari preparati da gruppi di
studenti su argomenti proposti dal docente e seminari tenuti dal docente
e/o suoi colleghi su argomenti specialistici e di approfondimento. Il corso
si propone di istruire lo studente sulla biochimica delle proteine e sulle
metodologie utilizzate per purificare proteine solubili e di membrana su
scala di laboratorio e industriale. Il corso si propone inoltre di fornire allo
studente informazioni riguardanti le applicazioni di varie classi di proteine
non catalitiche e di enzimi nel campo dell’industria alimentare,
farmaceutica e chimica.
The course consists of eight credits (64 hours). The course is
implemented both through classroom lectures using power point
presentations (seven credits) handed out to students, as well as through
seminars prepared by small groups of students on topics proposed by the
teacher, and seminars on specialized topics given by the teacher and/or
his colleagues (one credit).
The course aims to teach students about protein biochemistry and about
strategies for the purification of soluble and membrane proteins on
laboratory and industrial scale. Moreover, the course aims to teach
students about the application of non-catalytic proteins and enzymes in
the food, pharmaceutical, and chemical industries.
Alla fine dell’insegnamento lo studente dovrà conoscere la biochimica
delle proteine, i fattori che le stabilizzano/destabilizzano, le metodologie
utilizzate per preparare e purificare proteine di membrana, proteine non
catalitiche ed enzimi su scala di laboratorio e industriale. Gli studenti
dovranno inoltre conoscere le applicazioni di varie classi di proteine non
catalitiche e di enzimi nel campo dell’industria alimentare, farmaceutica
e chimica.
Lo studente dovrà essere in grado di progettare metodi di purificazione di
proteine di membrana, di proteine non catalitiche e di enzimi tenendo
conto della biochimica delle proteine, del grado di purificazione voluto,
dei costi e dell’uso previsto per la specifica proteina. Lo studente dovrà
inoltre essere in grado di ponderare possibili e potenziali applicazioni di
una specifica proteina in particolari settori dell’industria.
Gli studenti dovranno saper comunicare in modo chiaro e privo di
ambiguità le conoscenze e la ratio ad esse sottese. Le abilità
comunicative, la capacità di lavorare in gruppo, la capacità di analizzare
e sintetizzare informazioni, potranno essere sviluppate attraverso un ciclo
di seminari preparati da gruppi di studenti su argomenti proposti dal
docente. La preparazione dei seminari e la presentazione degli stessi in
aula sarà motivo di incitamento a interagire e comunicare in modo chiaro
con i colleghi studenti e con il docente.
At the end of the course, the students will be familiar with protein
biochemistry, with factors that stabilise/destabilise proteins, and methods
used to purify membrane proteins, non-catalytic proteins and enzymes
on laboratory and industrial scale. Moreover, the students will be familiar
with the applications of different classes of non-catalytic proteins and
enzymes in the food, pharmaceutical, and chemical industries.
The students should be able to plan purification methods for membrane
proteins, non-catalytic proteins and enzymes bearing in mind the protein
biochemistry, the wanted degree of purification, the costs, and the use of
a specific protein in a particular field of industry. Moreover, the students
should be able to ponder possible and potential applications of a specific
protein in a particular industrial sector.
The students should be able to communicate clearly the knowledge and
the related rationale. The communication ability, the capacity to work in
groups and the ability to analyse and summarise information will be
experienced through seminars prepared by small groups of students on
topics given by the teacher. The preparation of the seminars and their
presentation in the lecture room will encourage students to interact and
communicate between themselves and with the teacher.

Proteine globulari, fibrose e di membrana. Modifiche post-traduzionali.
Importanza delle catene laterali nella struttura e funzione delle proteine.
Folding, misfolding e denaturazione. Fattori destabilizzanti la struttura
delle proteine: detergenti, solventi organici, agenti ossidanti e caotropici,
temperatura, pH, congelamento, scongelamento. Stabilizzazione e
stoccaggio delle proteine: agenti stabilizzanti, refrigerazione,
liofilizzazione, conservazione e magazzinaggio.
Fonti di proteine: microbiche, animali, fungine e vegetali. Fonti da
organismi estremofili e da organismi geneticamente modificati. Screening
di nuovi biocatalizzatori.
Omogeneizzazione di tessuti e cellule su scala di laboratorio e su scala
industriale. Strategie per la purificazione di proteine solubili: Solubilità e
precipitazione frazionata al calore, mediante variazioni di pH e aggiunta
di solventi organici, salting-out e salting-in. Principali tecniche
cromatografiche per la purificazione di proteine su scala di laboratorio e
industriale.
Metodi di estrazione di proteine di membrana e loro ricostituzione in
liposomi.
Strategie di purificazione per proteine esocellulari e endocellulari di
interesse industriale, biomedico, farmaceutico ed analitico. Scale-up del
processo di estrazione e purificazione.
Proteine come corpi di inclusione: strategie di solubilizzazione e refolding.
Implicazioni tecniche ed economiche delle varie strategie di purificazione.
Enzimi e proteine per applicazioni industriali: enzimi immobilizzati,
tecniche di immobilizzazione, bioreattori. Biosensori: principi ed
applicazioni. Proteasi: classificazione ed usi industriali. Carboidrasi:
Applicazioni delle alfa-amilasi, beta-amilasi, glucoamilasi, alfa-(1-6)
glucosidasi, glucoso isomerasi. Enzimi degradanti la cellulosa,
l’emicellulosa, e pectina. Lipasi e loro applicazioni. Proteine del latte.
Enzimi e proteine di interesse biomedico, farmaceutico ed analitico.
Contaminanti proteici, virali, microbici, pirogenici. Prodotti del sangue,
enzimi ed anticorpi per scopi analitici e terapeutici, ormoni, fattori di
crescita, citochine, fattori di necrosi tumorale, interferoni, interleuchine.

and industrial applications.

Importance of side-chains on structure-function of proteins. Folding,
misfolding, denaturation. Destabilising factors of protein structure:
detergents, organic solvents, oxidant and chaotropic agents,
temperature, pH, freezing/thawing. Stabilisation and storage of proteins:
stabilising agents, cooling, freeze-drying, spray drying, sterilization,
preservation.
Protein sources: proteins from animals, plants, and from mesophilic and
extremophile organisms; protein production from genetically engineered
organisms. Screening of new enzymes. Homogenization of tissues and
cells on a laboratory and industrial scale.
Strategies for the purification of soluble proteins. Solubility and fractional
precipitation with temperature and pH changes, with the addition of
organic solvents, salting-out and salting-in. Main chromatographic
techniques for the purification of proteins on a laboratory and industrial
scale. Methods for purification of membrane proteins and for their
reconstitution into liposomes.
Purification of exocellular and endocellular proteins of industrial,
biomedical, pharmaceutical and analytical interest. Scale-up of the
protein extraction and purification process. Proteins as inclusion bodies:
solubilisation and refolding methods. Technical and economic
implications for different strategies of protein purification Proteins and enzymes for industrial applications: Immobilized enzymes,
immobilization techniques, bioreactors. Biosensors: principles and
applications. Proteases: classification and industrial uses. Carbohydrases:
applications of alpha-amylase, beta-amylase, glucoamylase, alpha-(1-6)
glucosidase, and glucose isomerase. Enzymes able to hydrolyse cellulose,
hemicellulose, and pectin. Lipases and their applications. Milk proteins.
Enzymes and proteins for medical, pharmacological, analytical, and food
applications. Microbial, viral, and pyrogenic contaminants. Blood
products, enzymes and antibodies for analytical and therapeutic
applications, hormones, growth factors, cytokines, tumoral necrosis
factors, interferons, interleukins.
Esame orale della durata media di 30 minuti.
L’esame orale è volto alla valutazione della conoscenza e comprensione
della biochimica delle proteine e degli altri argomenti di studio necessari
per risolvere i problemi relativi agli obiettivi del corso.
Il voto finale è attribuito in trentesimi. L’esame si intende superato
quando il voto è maggiore o uguale a 18. È prevista l’assegnazione del
massimo dei voti con lode (30 e lode).
Il voto finale viene attribuito sulla base delle risposte a 3 domande e sulla
base della qualità dei seminari presentati dagli studenti, che
contribuiranno al voto finale per un massimo di 2 punti. La lode viene
attribuita quando lo studente abbia dimostrato piena padronanza della
materia.
Oral examination of an average duration of 30 minutes.
The assessment method is an oral examination directed to evaluate the
knowledge and understanding of protein biochemistry and of the other
course topics necessary to solve problems related to the course
objectives.
The final mark is out of 30. Successful completion of the examination will
lead to grades ranging from 18 to 30, and 30 cum laude.
The final mark is attributed on the basis of the answers to three questions
and on the basis of the quality of the seminars given by the students. The
student seminars will contribute to the final mark for not more than two
points. The laude is attributed when the student demonstrates complete
mastery of the matter.
